RESUMO

Pasteurella pneumotropica is a bacterium that has so far not been described as a cause of placentitis in animals. Two cases of aborted equine fetuses were sent to the Department of Veterinary Pathology of the "Universidade Federal do Rio Grande do Sul" (SPV-UFRGS) for anatomopathological examination. Both cases presented suppurative placentitis associated with multiple basophilic bacterial cells. After bacterial isolation and biochemical analysis, P. pneumotropica was identified.

Dois casos de fetos equinos abortados foram enviados ao Setor de Patologia Veterinária da Universidade Federal do Rio Grande do Sul (SPV-UFRGS) para exame anatomopatológico. Em ambos os casos se observou placentite supurativa associada a múltiplas colônias bacterianas basofílicas. Após o isolamento bacteriano e análise bioquímica, indentificou-se P. pneumotropica.

Causes of abortion, stillbirth, and perinatal mortality in horses were investigated in the Department of Veterinary Pathology of the Federal University of Rio Grande do Sul (SPV-UFRGS) from 2000 to 2015. In this period, 107 cases were analyzed using macroscopic, microscopic, and complementary tests. Of these, 77 were aborted fetuses, 16 were stillbirths, and 14 were perinatal deaths. Conclusive diagnosis was established in 42.8% of the fetuses analyzed, with 28.6% classified as infectious origin, 9.1% as non-infectious, and 5.1% as other. Bacterial infections, especially those related to Streptococcus spp. were the most frequently observed. In stillborn foals, diagnosis was established in 62.5% of cases, and 50% of these were related to non-infectious causes, such as dystocia and birth traumas. As for perinatal mortality, a conclusive diagnosis was reached in 78.57% of cases, and infectious causes associated with bacterial infections accounted for 64.1% of these diagnoses.


Causas de aborto, natimortalidade e mortalidade perinatal em equinos foram investigadas no Setor de Patologia Veterinária da Universidade Federal do Rio Grande do Sul (SPV-UFRGS) durante o período de 2000 a 2015. Nesse período, foram analisados 107 casos através de exames macroscópico, microscópico e exames complementares, desses 77 correspondiam a fetos abortados, 16 natimortos e 14 mortes perinatais. Diagnóstico conclusivo foi estabelecido em 42,8% dos fetos analisados e classificados como origem infecciosa em 28,6% dos casos, não infecciosa com 9,1% e outros com 5,1% dos casos. As infecções bacterianas, em especial as relacionadas a Streptococcus spp. foram as mais frequentemente observadas. Em potros natimortos, diagnostico foi estabelecido em 62,5% dos casos, e destes, 50% foram relacionados a causas não infecciosas, como distocia e traumas durante o parto. Quanto a mortalidade perinatal, em 78,57% dos casos houve um diagnostico conclusivo, e as causas infecciosas associadas a infecções bacterianas corresponderam a 64,1% desses diagnósticos.

Mastitis may be caused by a wide range of microorganisms able to induce distinct lesions in mammary tissues. This study aims to characterize the gross and microscopic features of mastitis in dairy cows and to correlate them with the pathogens involved. The udders of slaughtered dairy cows were inspected and milk samples from each mammary quarter or fragments of the parenchyma were sent for microbiological analysis, and tissue collected for histopathological evaluation. A total of 148 cows and 592 mammary quarters were collected. From these, 432 quarters (73%) had mastitis and in 160 (27%), no changes were observed. Mastitis was classified into seven patterns based on the histopathological findings, of which mixed, lymphoplasmacytic, and suppurative mastitides were the most prevalent with 35.9% (155/432), 27.1% (117/432), and 14.3% (62/432) of the cases, respectively. These patterns were associated with the same set of pathogens: Streptococcus spp., coagulase-negative Staphylococcus (CNS), Staphylococcus aureus, Streptococcus agalactiae, Streptococcus uberis, and Corynebacterium bovis. The pyogranulomatous pattern represented 7.2% (31/432) of the cases with distinct distribution based on the agent involved, mostly S. aureus and Nocardia sp. Abscedative mastitis accounted for 6.0% (26/432) of the cases; it was characterized by multiple abscesses in the parenchyma and was mainly caused by Trueperella pyogenes. Necrosuppurative mastitis represented 5.8% (25/432) of the cases which were characterized by severe parenchyma necrosis and were caused by bacteria such as CNS and Escherichia coli. The granulomatous pattern represented 3.7% (16/432) of the cases and was occasionally associated with Mycobacterium sp.

Southern right whales Eubalaena australis (SRW) use the southern coast of Brazil as a wintering and calving ground. Other than anthropogenic threats, there is limited knowledge on health and disease aspects for this species. We report the gross and microscopic findings and microbiological identification of streptococcal septicemia in a SRW calf. Main gross findings included fibrinosuppurative omphalitis and urachocystitis, suppurative cystitis, valvular endocarditis and myocarditis, embolic pneumonia, suppurative myositis and osteoarthritis, and lymphadenomegaly. Histological examination confirmed the above inflammatory processes and indicated disseminated Gram-positive coccoid septicemia. PCR analysis, based on the 16S rRNA gene from bacteria isolated on blood agar, identified Streptococcus dysgalactiae. Pathologic and microbiologic analysis indicated that ß-hemolytic S. dysgalactiae septicemia, presumably initiated as ascending omphalic infection, was responsible for stranding and death in this individual. These results further confirm pathogenicity of streptococci in cetaceans and add to the limited health and disease related pathology knowledge for this species.

The diagnosis of the cause of death in goats submitted to necropsy from January 2000 to December 2016 by Setor de Patologia Veterinária from the Universidade Federal do Rio Grande do Sul was reviewed. Epidemiological features, such as the breed, sex and age, in addition to the clinical and pathological features were evaluated. During this period, 322 goats were necropsied, in which a conclusive diagnosis was obtained in 290 (90%) goats. Goats that were part of other experimental study were excluded from this study. From these 290 cases, 167 (57.6%) corresponded to diseases of infectious origin and toxinfectious diseases, while 123 (42.4%) were classified as non-infectious conditions. Infectious diseases included 55 cases of bacterial origin, 59 cases with parasitary involvement, 14 cases of viral origin, and 39 toxinfectious cases. Non-infectious conditions were grouped into metabolic diseases (44 cases), plants or chemical substances poisoning (36), mineral and nutritional deficiencies (20), and neoplasms and developmental disorders (5). In the remaining 18 cases, a conclusive diagnosis was obtained, however the conditions did not fit into those criteria and were classified as "others". The age range of the goats in this study was from 1 day-old to 10 years-old. Most of the goats were females (201), while 121 were males. Affected breeds included Boer, Saanen, Anglo-Nubian, Toggenburg and mixed breeds. Parasitic, infectious and toxin-infectious diseases were the main cause of deaths, especially haemonchosis, pleuropneumonia, eimeriosis and enterotoxemia. Among the non-infectious conditions, metabolic disorders, especially rumen acidosis, pregnancy toxemia and urolithiasis, were directly related to the management employed in the property. Plant poisoning diagnosis was also highlighted with locally present plants, such as Sida carpinifolia, as the most important.(AU)


Foram revisadas as causas de morte de caprinos submetidos ao exame de necropsia no período compreendido entre janeiro de 2000 e dezembro de 2016 pelo Setor de Patologia Veterinária da Universidade Federal do Estado do Rio Grande do Sul, Brasil. Avaliaram-se os dados epidemiológicos relacionados à raça, sexo e idade, assim como as alterações macroscópicas e microscópicas observadas. Durante esse período, foram recebidos para necropsia 322 caprinos, nos quais em 290 casos (90%) foi obtido o diagnóstico conclusivo. Caprinos destinados à experimentação animal foram excluídos deste estudo. Do total de 290 casos, 167 (57,6%) corresponderam a enfermidades de origem infecciosa e toxi-infecciosa, enquanto 123 (42,4%) em causas não infecciosas. Entre as doenças infecciosas foram contabilizados 55 casos de origem bacteriana, 59 casos com envolvimento parasitário, 14 casos de origem viral, e 39 casos toxi-infecciosos. As doenças de caráter não infeccioso foram agrupadas em doenças metabólicas (44 casos), intoxicações por plantas ou substâncias tóxicas (36), deficiências minerais e nutricionais (20), neoplasias e distúrbios no desenvolvimento (5). Dezoito casos com diagnóstico conclusivo, mas que não se enquadravam nestas classificações foram agrupadas como "outros". A faixa etária dos caprinos neste estudo variou de um dia a 10 anos. A maior parte dos animais eram fêmeas (201), enquanto 121 eram machos. As raças dos caprinos necropsiados neste período incluíram Boer, Saanen, Anglo-Nubiano, Toggenburg e sem raça definida (SRD). As enfermidades de origem parasitárias, infecciosas e toxi-infecciosas, especialmente hemoncose, eimeriose, pleuropneumonias e a enterotoxemia acarretaram o maior número de mortes. Das causas não infecciosas, distúrbios metabólicos, como acidose lática ruminal, toxemia da prenhez e urolitíase, estiveram diretamente relacionadas com o manejo empregado nas propriedades. Evidenciou-se também a importância do diagnóstico de intoxicações por plantas relevantes da região, como Sida carpinifolia.(AU)

Respiratory diseases cause significant veterinary costs, reduce performance and require withdrawal of horses. Yet, studies of the causes of pneumonia in horses are scant. This study aimed to describe the pathological and microbiological features of lung lesions in slaughtered horses in southern Brazil. In this study, 84 samples of lungs were examined, and a conclusive diagnosis was obtained in 74 cases. These were composed of bronchopneumonia in 50 cases, followed by granulomatous eosinophilic pneumonia (9/74), recurrent airway obstruction (7/74), lung fibrosis (4/74), lung hemorrhage (3/74) and pulmonary pythiosis (1/74). Bronchopneumonia had grossly firm focally extensive yellow to dark-red areas, which consisted microscopically of multifocal to coalescing infiltrate of degenerate neutrophils. Streptococcus equi subsp. zooepidemicus was identified in 21 of the 50 cases. Granulomatous eosinophilic pneumonia had multifocal pinpoint firm-hard yellow areas, which microscopically were composed of granulomas with a mineralized center surrounded by collagen fibers and severe infiltrate of eosinophils. Recurrent airway obstruction had mild multifocal pinpoint firm white areas that consisted microscopically of large amounts of mucus inside bronchi and bronchiole. Lung fibrosis had two patterns: focally extensive areas of consolidation and firm nodular areas. Microscopically, the first pattern had interstitial to peribronchial fibrosis, while the second had, in addition to the interstitial fibrosis, a severe pneumocyte hyperplasia and an alveolar infiltrate of neutrophils and macrophages with rare intranuclear inclusion bodies (equine herpesvirus 5, EHV-5). Pulmonary pythiosis presented a focal firm nodular area, with multiple kunkers observed in the cut surface, which corresponded microscopically to areas of necrosis surrounded by a mixed inflammatory infiltrate. At the periphery of the necrotic areas, multiple negatively stained hyphae were observed, which were evidenced through Grocott's stain and immunohistochemistry anti-Pythium insidiosum.(AU)


Doenças respiratórias causam em equinos custos significativos com tratamento veterinário, redução de performance e descarte de animais. No entanto, estudos que abordem as causas de pneumonia em equinos são escassos. O objetivo deste estudo foi descrever os aspectos patológicos e microbiológicos de lesões pulmonares em equinos abatidos em matadouro-frigorífico no Sul do Brasil. Neste estudo, 84 amostras de pulmões foram examinadas, e o diagnóstico conclusivo das condições foi obtido em 74 casos. Esses foram compostos por broncopneumonia em 50 casos, seguido por pneumonia granulomatosa eosinofílica (9/74), obstrução aérea recorrente (7/74), fibrose pulmonar (4/74), hemorragia pulmonar (3/74) e pitiose pulmonar (1/74). A broncopneumonia era caracterizada macroscopicamente por áreas focalmente extensas firmes de coloração amarelada a vermelho-escuras, as quais consistiam microscopicamente em infiltrado multifocal a coalescente de neutrófilos degenerados. Streptococcus equi subsp. zooepidemicus foi identificado em 21 dos 50 casos. A pneumonia eosinofílica granulomatosa era caracterizada por áreas multifocais puntiformes firmes a duras e amareladas, que microscopicamente eram compostas por granulomas com área central mineralizada circundados por fibras de colágeno e infiltrado acentuado de eosinófilos. A obstrução aérea recorrente era caracterizada por discretas áreas puntiformes firmes e brancacentas que consistiam microscopicamente em grande quantidade de muco no interior de brônquios e bronquíolos. A fibrose pulmonar exibia dois padrões: áreas de consolidação focalmente extensas e áreas nodulares firmes. Microscopicamente, o primeiro padrão exibia fibrose intersticial a peribronquial, enquanto no segundo padrão havia, além da fibrose intersticial, intensa hiperplasia de pneumócitos e infiltrado alveolar de neutrófilos e macrófagos com raros corpúsculos de inclusão intranucleares (herpesvírus equino 5, EHV-5). A pitiose pulmonar exibia uma área nodular firme focal com múltiplos kunkers ao corte, os quais correspondiam microscopicamente a áreas de necrose circundadas por infiltrado inflamatório misto. À periferia das áreas necróticas, múltiplas imagens negativas de hifas eram observadas, as quais foram evidenciadas através da coloração de Grocott e imuno-histoquímica anti-Pythium insidiosum.(AU)

Johne's disease is a chronic granulomatous enteritis caused by Mycobacterium avium subsp. paratubercolosis (MAP) which affects ruminants worldwide and has a significant economic impact. MAP has also been associated with human Crohn's disease, although this connection is not well established. MAP is highly adapted for survival within host macrophages and prevents macrophage activation, blocks phagosome acidification and maturation, and attenuates presentation of antigens to the immune system. The consequence is a very long silent infection before clinical signs are observed. The present work examined the transcriptome of bovine monocyte-derived macrophages (MDM) infected with the L1 strain of MAP at 2h, 6h and 24h post infection using RNA-seq. Pathway over-representation analysis of genes differentially expressed between infected vs. control MDM identified that immune related pathways were affected. Genes belonging to the cytokine-cytokine receptor interaction pathway and members of the JAK-STAT pathway, which is involved in the regulation of immune response, were up-regulated. However, in parallel inhibitors of immune functions were activated, including suppressor of cytokine signaling (SOCS) and cytokine-inducible SH2-containing protein (CISH), which most likely suppresses IFNγ and the JAK/STAT signaling cascade in infected MDM, which may favour MAP survival. After exposure, macrophages phagocytise pathogens, activate the complement cascade and the adaptive immune system through the antigen presentation process. However, data presented here suggest that genes related to phagocytosis and lysosome function are down regulated in MAP infected MDM. Genes of MHC class II and complement pathway were also down-regulated. This study therefore shows that MAP infection is associated with changes in expression of genes related to the host immune response that may affect its ability to survive and multiply inside the host cell.

This study describes suppurative infectious diseases of the central nervous system (CNS) in domestic ruminants of southern Brazil. Reports from 3.274 cattle, 596 sheep and 391 goats were reviewed, of which 219 cattle, 21 sheep and 7 goats were diagnosed with central nervous system inflammatory diseases. Suppurative infectious diseases of the CNS corresponded to 54 cases (28 cattle, 19 sheep and 7 goats). The conditions observed consisted of listerial meningoencephalitis (8 sheep, 5 goats and 4 cattle), suppurative leptomeningitis and meningoencephalitis (14 cattle, 2 goats and 1 sheep), cerebral (6 cattle and 2 sheep), and spinal cord (7 sheep) abscesses, and basilar empyema (4 cattle and 1 sheep). Bacterial culture identified Listeria monocytogenes (9/54 cases), Escherichia coli (7/54 cases), Trueperella pyogenes (6/54 cases) and Proteus mirabilis (1/54 cases). All cases diagnosed as listeriosis through histopathology yielded positive immunostaining on immunohistochemistry, while 12/17 of the cases of suppurative leptomeningitis and meningoencephalitis presented positive immunostaining for Escherichia coli. Meningoencephalitis by L. monocytogenes was the main neurological disease in sheep and goats, followed by spinal cord abscesses in sheep. In cattle, leptomeningitis and suppurative meningoencephalitis was the most frequent neurological disease for the species, and E. coli was the main cause of these lesions. Basilar empyema, mainly diagnosed in cattle, is related to traumatic injuries, mainly in the nasal cavity, and the main etiologic agent was T. pyogenes.(AU)


Neste trabalho são descritas as doenças neurológicas infecciosas supurativas de ruminantes domésticos na Região Sul do Brasil. Foram avaliados laudos de 3.274 bovinos, 596 ovinos e 391 caprinos, dos quais 219 bovinos, 21 ovinos e sete caprinos foram diagnosticados como doenças inflamatórias no sistema nervoso central. As doenças neurológicas infecciosas supurativas corresponderam a 54 casos (28 bovinos, 19 ovinos e sete caprinos). As enfermidades observadas foram meningoencefalite por Listeria monocytogenes (oito ovinos, cinco caprinos e quatro bovinos), leptomeningite e meningoencefalite supurativa (14 bovinos, dois caprinos e um ovino), abscessos cerebrais (seis bovinos e dois ovinos) e medulares (sete ovinos); e empiema basilar (quatro bovinos e um ovino). Através do isolamento bacteriano foram identificados: L. monocytogenes (9/54 casos), Echerichia coli (7/54 casos), Trueperella pyogenes (6/54 casos) e Proteus mirabilis (1/54 casos). Todos os casos diagnosticados como listeriose por histologia foram positivos na imuno-histoquímica para L. monocytogenes, e 12/17 casos de leptomeningite e meningoencefalite supurativa foram positivos na imuno-histoquímica para E. coli. A meningoencefalite por L. monocytogenes representou a principal enfermidade neurológica em ovinos e caprinos, seguido dos abscessos medulares em ovinos. A leptomeningite e meningoencefalite supurativa foi a doença neurológica supurativa mais frequente em bovinos e o principal agente causador da lesão foi E. coli. O empiema basilar, frequentemente, diagnosticado em bovinos, foi relacionado com lesões traumáticas, principalmente, de cavidade nasal e o principal agente causador foi T. pyogenes.(AU)

The biofilm-associated protein (Bap) of Staphylococcus aureus is a high molecular weight cell-wall-anchored protein involved in biofilm formation, first described in bovine mastitis strains from Spain. So far, studies regarding Bap were mainly based on the Spanish strain V329 and its mutants, but no information on the genetic variability of bap-positive Staph. aureus strains is yet available in the literature. The present study investigated the molecular characteristics of 8 bap-positive Staph. aureus strains from subclinical bovine mastitis, isolated in 5 herds; somatic cell counts (SCC) of milk samples were also registered. Strains were characterised using MLST, SPA typing and microarray and the results were compared with V329. All isolates from this study and V329 were assigned to ST126, t605, but some molecular differences were observed. Only herd A and B strains harboured the genes for ß-lactams resistance; the leukocidin D/E gene, a type I site-specific deoxyribonuclease subunit, 3rd locus gene and serin-protease A and B were carried by all strains, but not by V329, while serin-protease E was absent in V329 and in another isolate. Four isolates and V329 harboured the fibronectin-binding protein B gene. SCC showed the highest value in the milk sample affected by the only strain carrying all the virulence factors considered. Potential large variability of virulence was evidenced among V329 and all bap-positive Staph. aureus strains considered: the carriage of fnb could enhance the accumulation of biofilm, but the lack of lukD/E and splA, B or E might decrease the invasiveness of strain.

Staphylococcus aureus isolates from dairy cow mastitis are not always consistent with the characteristic morphology described, and molecular investigation is often needed. The aim of the study was to develop a duplex real-time PCR assay for rapid identification of Staph. aureus isolates, targeting both nuc and Sa442. Overall, 140 isolates collected from dairy cow mastitis in 90 different herds, were tested. All strains had been identified using morphological and biochemical characteristics. DNA from each strain was amplified in real-time PCR assay, to detect nuc or Sa442. Thereafter, a duplex real-time PCR assay was performed, and specificity of the amplified products was assessed by high resolution melting curve analysis. Out of 124 Staph. aureus isolates, 33 did not show the typical morphology or enzymic activity; in 118 strains, the two melt-curve peaks consistent with nuc and Sa442 were revealed, while 2 isolates showed only the peak consistent with Sa442. Four isolates bacteriologically identified as Staph. aureus, were PCR-negative and were further identified as Staph. pseudintermedius by sequencing. Staph. pseudintermedius and coagulase-negative staphylococci did not carry nuc or Sa442. The results showed the correct identification of all isolates, comprehending also coagulase-or nuc-negative Staph. aureus, while other coagulase-positive Staphylococci were correctly identified as non-Staph. aureus. Both sensitivity and specificity were 100%. High resolution melting analysis allowed easy detection of unspecific products. Finally, the duplex real-time PCR was applied directly to 40 milk samples, to detect infected mammary quarters. The assay confirmed the results of bacteriological analysis, on Staph. aureus-positive or-negative samples. Therefore, the proposed duplex real-time PCR could be used in laboratory routine as a cost-effective and powerful tool for high-throughput identification of atypical Staph. aureus isolates causing dairy cow mastitis. Also, it could be applied directly to milk samples, to detect Staph. aureus mammary infections avoiding bacteriological analysis.

Yersinia enterocolitica é uma bactéria Gram-negativa que causa infecções em diversas espécies de mamíferos. O agente, geralmente, provoca infecções restritas ao intestino e linfonodos mesentéricos, porém a infecção pode se tornar sistêmica ocasionando lesões em outros órgãos como fígado e baço. Neste trabalho descrevem-se dois surtos de infecções sistêmicas causadas pela Yersinia enterocolitica em criatórios comerciais de chinchilas no Rio Grande do Sul (Brasil). Os proprietários relatavam que os animais acometidos apresentavam apatia, anorexia e morte. Foram encaminhados 13 animais para a realização de necropsia. No exame post mortem dos animais observou-se esplenomegalia, hepatomegalia e áreas multifocais esbranquiçadas no fígado, baço, pulmões, rins e intestino. No exame microscópico visualizou-se infiltrado inflamatório de neutrófilos e macrófagos, necrose, deposição de fibrina e ocasionalmente pode ser observado coco-bacilos no centro das áreas de necrose. No cultivo bacteriológico obteve-se o crescimento de Yersinia enterocolitica nos animais provenientes dos dois criatórios. O agente foi isolado de amostras no fígado, baço, intestino e pulmões dos animais necropsiados, além do cultivo de fezes de animais de uma das propriedades acometidas. A yersiniose, portanto, é uma patologia que deve ser investigada em casos de mortalidade de chinchilas.


Yersinia enterocolitica is a Gram-negative bacterium, which causes infections in several mammalian species. It is often recognized as an agent causing intestinal and mesenteric lymph nodes lesions. However, Yersinia enterocolitica infection may also become systemic, with lesions in others organs such as liver and spleen. This paper describes outbreaks of systemic infection due to Yersinia enterocolitica in two commercial chinchilla breeders in Rio Grande do Sul (Brazil). Owners reported that affected animals showed apathy, anorexia prior to death. Macroscopic examination performed in 13 animals revealed splenomegaly, hepatomegaly and multifocal whitish pinpoint foci in liver, spleen, lung, kidney and intestine. Microscopically, the affected tissues had infiltration of neutrophils and macrophages, as well as fibrin and necrosis with central areas containing cocobacilli bacteria. Yersinia enterocolitica was isolated from liver, spleen, lung and intestine samples from animals of both breeders, and from feces of chinchillas of one of the breeders. Therefore, yersiniosis is a disease to be investigated in cases of mortality of chinchillas.
